Not really missable , but you will not be able to get it until after the Annihilator boss fight, after some story related stuff and scenes you will be back in the Keystone realm , in a dungeon / tunnel there will be a portal directly in front of you, enter here and fight the 3 waves of enemies. These should include the last enemies you need for the achievement.
The 3 you need will be here but they will also appear later after anonther boss and you progress with the story.
