To unlock Colossus, go to the X-Mansion. In the right side of the courtyard, a Sentinel will appear. Defeat him and Colossus will be unlocked (150,000 studs to purchase). Then simply set one character as him and the other as Wolverine; take control of Colossus and press Square towards Wolverine until he throws him, this may take multiple tries before he performs the move.
